The DynamicOrgan® O2-Sensor Kit, developed in collaboration with PyroScience allows for real-time, contact-less, non-invasive detection of oxygen consumption inside an organ model such as lung, gut or liver-on-chip.

The heart of DynamicOrgan® O2-Sensor Kit is the 2-channel biochip (BC002) with oxygen sensor spots from PyroScience.
The optical sensors from PyroScience are biocompatible and therefore ideal for sensitive biological methods, such as organ-on-chip. The oxygen meter (FirePlate) excites the oxygen sensitive indicator with its unique REDFLASH technology and detects the oxygen dependent luminescence emission. The higher the oxygen content, the more quenching of the luminescent signal is taking place, allowing for a precise calculation of the oxygen content in the channel of the biochip.
Three sensor spots are incoperated into our biochip, enabling oxygen measurement in the top and bottom channel.
The top channel features a single sensor spot in the center of the top foil for general oxygen level measurement inside the channel. The bottom channel includes two sensor spots on the bottom foil: one positioned at the inlet and another at the outlet. This enables the calculation of oxygen consumption by the cells incorporated in the bottom channel.

This measurement of oxygen levels inside the biochip, without direct intervention in the medium, is a particular advantage of the system. It allows real-time recording of changes in oxygen and its consumption by cells without compromising the integrity of the tissues or organ model.
The sensor materials are robust against various sterilization methods, including 70% ethanol, ethylene oxide, and plasma irradiation. This makes them easier to use in regulated and sterile laboratory environments, such as incubators.
The REDFLASH technology uses an oxygen-sensitive indicator excitable with orange-red light (610–630 nm) and emitting in the near infrared (760–790 nm). It offers fast response times and low cross-sensitivity. The red excitation minimizes autofluorescence and stress in biological systems.
Oxygen Sensors are currently available for our 2-channel biochip (BC002) only. Any organ models previously developed on this biochip can be transferred to the O2-Sensor biochip.

In addition to the kit, you will require a peristaltic pump and the measurement device for your oxygen sensor read-outs (FirePlate from PyroScience) plus a holding frame suitable to host 3 biochips in parallel. Should you already have a preexisting Fireplate or pump system only the holding frame specifically designed to hold 3 biochips will be needed in addition to the kit.
The bottom channel of the O2-Sensor biochips is equipped with a sensor spot at the inlet as well as the outlet. This enables the calculation of oxygen consumption by the tissue incorporated in the bottom channel.
