This kit contains all consumables needed to combine a barrier with spheroids or organoids in one multilayer organ model. This could be used to combine a vasculature with a tumor or other spheroids of your choice.

The Spheroid Kit hosts our BC003, a biochip containing two membranes (1x porous, 1x microcavity) dividing the culture chamber into a top, middle and bottom channel accessible via ports. The middle channel with the microcavity membrane can host up to 25 spheroids or organoids.

Cell suspensions and coatings can be introduced individually into each channel, enabling the creation of cell layers and complex tissues on both sides of the membrane.

 

The separation of the channels allows nourishing of the tissues and spheroids with media and individual flow rates per channel (or no flow at all).

 

Additionally, circulating cell populations can be introduced. Matching reservoirs facilitate perfusion and ensure nutritional supply up to three days without medium exchange.

Additionally, all kits are available with different membranes (0.4, 3, 5 or 8µm) and microscope cover glass at the bottom.

What do I need to get started?

In addition to the kit, you will require a peristaltic pump. If you don’t have one, you can order a DynamicOrgan® System and will receive a peristaltic pump in addition to the Spheroid Kit.

What is a spheroid-on-chip model?

Spheroid-on-chip combines advanced 3D cell culture models like spheroids and organoids with microfluidic on-chip devices.

What advantage does spheroid-on-chip offer compared to using spheroids in static culture?

Traditional methods of spheroid formation, such as hanging drop cultures or microwell plates, have been instrumental in the development of 3D cell culture. However, they have significant limitations. The static nature of these culture environments can lead to rapid depletion of oxygen and nutrients, as well as accumulation of waste products, which adversely affect spheroid viability and the reliability of experimental results. In contrast, microfluidic spheroid-on-chip models offer a more sophisticated approach to spheroid culture. These biochips allow for precise control over the microenvironment, including regulation of chemical gradients, pressure, and shear stress. This level of control is crucial for mimicking the dynamic conditions of in vivo tissues.

How is spheroid-on-chip in our Spheroid Kit different to using spheroids in our 3-Channel Kit?

The Spheroid Kit includes the BC003 biochip hosting one flat and one micro-cavity membrane. This allows for arrangement of spheroid into a single layer and for easier imaging and sampling. The 3-Channel Kit includes the BC003 biochip with 2 flat membranes. Here, spheroids are inserted together with a gel matrix which allows the spheroids to arrange in a random fashion closer to the in vivo situation of a tumor.

What is the maximum size of spheroids that can be introduced in the biochip?

Due to the channel size, spheroid diameter is limited to a maximum of 800µm.

Can Spheroids be recovered during or after the experiment?

With the spheroid kit spheroids or organoids are loaded without a gel matrix onto the biochip allowing for them to be recovered more easily. Spheroids can either be recovered from the channel directly or through cutting of the membrane after the experiment has concluded.
